Microtomes
Microtomes are cutting devices designed to prepare samples for observa
tionMicrotomes for the preparation of the microscopy under a microscope. To achieve the standards such samples require, the microtomes are able to cut extremely thin slices of the specimen. Usually modern microtomes create slices with a thickness of 0.1 to 100 m. For comparison: a human hair has an average thickness of 50 to 70 m. The history of the microtomes begins approximately at the same time as of light microscopy. To observe those samples it is necessary to create sections that are thin enough to be transmitted by light. The first prototypes were microtomes with basically simple blades (mostly razor blades), which were built to cut sections hand operated. With increasing requirements on samples the microtomes began to develop. The primal microtomes in present day sense were developed in 1770. With those the specimen could be fixed and the desired thickness of the sections could be adjusted by meanings of screws. Today mechanic microtomes consist of a knife-block, a specimen holder and an advancement mechanism. The quality of the samples depends on the type of advancement mechanism, the geometric design of the knife and the declination (angle of contact between the sample vertical and knife blade). Furthermore the result can be affected by the prior preparation of the sample (e.g. cryosectioning technique). Aside from mechanic microtomes today the use of laser microtomes is increasing, which allow contact free slicing. Following detailed descriptions on the operating mode of microtomes:

Sledge microtomes
These microtomes consist of a fixed holder for the samples and a knife affixed to a movable sledge. To achieve a stable cut, the sledges of those microtomes are often relatively heavy. During the slicing process the knife is being pressed through the sample. With sledge microtomes a cut thickness of 1 to 60
m can be realized.

Rotary microtomes

Those microtomes, which are also called minot microtomes, have an affixed knife and a moving sample holder. The rotary microtomes got their name due to a flywheel impelling the motion of the sample holder, except the rotary motion of the flywheel is converted into a linear horizontal motion. Commonly the sample holder of those microtomes performs a downward motion towards the knife. The created sample cuttings are then gathered on the blade. These microtomes hold the advantage of compensating varying hardness of the specimen due to a heavy mass of the flywheel resulting in even sections. Rotary microtomes enable the creation of sections with a cut thickness of 1 to 60
m.

Cryo - microtomes
Cryo - microtomes are a special type of rotary microtomes. Therefore the specimen is held in a deep-frozen container, usually cooled by liquid nitrogen. The reduced temperature allows for the hardness of those specimens to be increased. 

Laser microtomes
Laser- microtomes use a special laser to create the sections. That so called ultra short pulse laser stands out due to a strong focusing and very short pulse intervals. As a result extremely thin sections can be created without causing thermal harm to the specimen material. These microtomes allow creating samples with thickness between 10 and 100
m.
